Swim-up

Se denomina Swim-up a la técnica utilizada para capacitar los espermatozoides in vitro. La muestra de semen se centrifuga (unos 10 min a 400g), los espermatozoides forman el pellet. Se decanta y se añade de 0,3 a 0,5 ml de medio de cultivo procurando que resbale suavemente por las paredes del tubo, para evitar que se formen burbujas. Se incuba durante 45 min. En este tiempo solo los espermatozoides con mejor movilidad colonizarán la parte superior del medio y son los que serán rescatados. Las principales ventajas de esta técnica de capacitación es que es fácil de realizar, muy barata y la fracción resultante es muy limpia. Sin embargo, presenta una baja tasa de recuperación de espermatozoides, por lo que esta estrategia está indicada para muestras de semen de elevada concentración y movilidad. [1]

Esta técnica, actualmente, se continúa utilizando aún en los laboratorios FIV. Resulta útil en normozoospermia. Se consiguen fracciones con más del 90% de espermatozoides con calidad A+B y los citados espermatozoides se encuentran en contacto con el debris celular.[2]
